IGSF1deficiency is caused by a defect in the IGF1gene which is located on the X chromosome. Conditions of the X-chromosome affect boys more severely than girls.
The exact mechanism by which IGSF1deficiency causes central hypothyroidism and macroorchidism is still not completely clear.
