Loss of function variants in RFX3 result in a neurobehavioral phenotype that includes intellectual disability of varying severity (borderline to moderate), autism or autistic features, and disruptive behaviors. 18 cases with 15 distinct variants have been reported thus far. There is one family with an inherited variant; all others are de novo.
